Spectral Distribution of one-dimensional Photonic Quasicrystals: The Role of Irrational Numbers

In this paper, we construct a one-dimensional photonic quasicrystal by combining two incommensurate spatial harmonics, where the ratio of their periods is the irrational number \beta. We evaluate the photonic quasicrystal accurately by a generalized spectral method that embeds the quasiperiodic structure into a higher-dimensional periodic system. We study the spectral distribution of one-dimensional photonic quasicrystals and find some interesting phenomena. As the computational resolution N increases, there are more eigenvalues within finite frequency bandwidths, and the maximum localization always occurs at spectral gap edges for states near index N + 1. By varying \beta within the range of (0,1), we present a butterfly-shaped spectral structure with abundant band gaps. We find that the spectral structure factor Q (defined as I_{mg}/N, where I_{mg} is the maximum gap index) exhibits different linear patterns as \beta changes: Q = 1 - \beta when \beta < \beta c, while Q = \beta when \beta > \beta c, where \beta c \approx 0.424 is the transition point. This linear relationship holds robustly in the strong quasiperiodic regime (\beta away from 0 or 1) and is independent of the specific type of irrational number used. The relationship disappears (weak quasiperiodic regime) near \beta = 0 or \beta = 1. It demonstrates that the intrinsic spectral properties of one-dimensional photonic quasicrystals are fundamentally governed by the magnitude of the irrational parameter \beta.

Integration of Efficacy Biomarkers Together with Toxicity Endpoints in Immune-Oncology Dose Finding Studies

The primary objective of phase I oncology studies is to establish the safety profile of a new treatment and determine the maximum tolerated dose (MTD). This is motivated by the development of cytotoxic agents based on the underlying assumption that the higher the dose, the greater the likelihood of efficacy and toxicity. However, evidence from the recent development of cancer immunotherapies that aim to stimulate patients' immune systems to fight cancer challenges this assumption, particularly further escalation after a certain dose level might not necessarily increase the efficacy. Dose escalation study of molecular targeted agents (MTA) often does not only rely on the safety profile. In this paper, we propose a simple and flexible model that uses multivariate Gaussian latent variables to integrate toxicity endpoint and efficacy biomarker. This model can be easily extended to incorporate additional immune biomarkers. By simultaneously considering multiple outcomes, the proposed method is better at identifying the biologically optimal dose, which results in better decision-making. Simulation studies showed that the proposed method has desirable operating characteristics by determining the target dose with an optimal risk-benefit trade-off. We have also implemented our proposed method in a user-friendly R Shiny tool.

Statistical Issues and Recommendations for Clinical Trials Conducted During the COVID-19 Pandemic

The COVID-19 pandemic has had and continues to have major impacts on planned and ongoing clinical trials. Its effects on trial data create multiple potential statistical issues. The scale of impact is unprecedented, but when viewed individually, many of the issues are well defined and feasible to address. A number of strategies and recommendations are put forward to assess and address issues related to estimands, missing data, validity and modifications of statistical analysis methods, need for additional analyses, ability to meet objectives and overall trial interpretability.
