Counterion Effects on Nano-confined Metal-Drug-DNA Complexes

We have explored morphology of DNA molecules bound with Cu-complexes of piroxicam molecules, a non-steroidal anti-inflammatory drug (NSAID), under one-dimensional confinement of thin films and have studied the effect of counterions present in a buffer. X-ray reflectivity at and away from the Cu K absorption edge and atomic force microscopy studies reveal that confinement segregates the drug molecules preferentially in a top layer of the DNA film, and counterions enhance this segregation.

Effects of Counterions on Nano-confined DNA Thin Films

DNA molecules spin-coated on amorphous quartz substrates are shown to form stable films. Electron density profiles (EDPs) along the film depth show that film prepared from aqueous solution of DNA exhibits layering of the molecules in three stacks parallel to the substrate whereas film prepared from counterion added solution does not have layering but have smaller thickness and enhanced surface roughness, although both films have 'liquid-like' height-height correlations. We explain these results by a model of film of a 'liquid' comprising of rod-like molecules where the counterion concentration in the liquid determines the rod length.

Onion structures induced by hydrophilic and hydrophobic ions in a binary mixture

Phase transition is observed between one-phase disordered phase and an ordered phase with multi-lamellar (onion) structures in an off-critical mixture of D$_2$O and 3-methylpyridine (3MP) containing a salt at 85mM. The salt consists of hydrophilic cations and hydrophobic anions, which interact asymmetrically with the solvent composition fluctuations inducing mesophases. The structure factor of the composition distribution obtained from small-angle neutron scattering has a peak at an intermediate wave number in the disordered phase and multiple peaks in the ordered phase. Lamellar layers forming onions are composed of solvation-induced charged membranes swollen by D$_2$O. The onion phase is realized only for small volume fractions of 3MP (in D$_2$O-rich solvent).

Smooth/rough layering in liquid-crystalline/gel state of dry phospholipid film, in relation to its ability to generate giant vesicles

Morphological changes in a dry phospholipid film on a solid substrate were studied below and above the main transition temperature, between the gel and liquid-crystalline phases by phase-contrast microscopy and AFM. A Phospholipid film in the liquid-crystalline phase exhibits flat, smooth layering, whereas that in the gel phase shows rough, random layering. These film morphologies are discussed in relation to the ability to form giant vesicles through the natural swelling method.
