Energy-Stable Parametric Operator Inference across Regime Transitions: Application to Unconfined Buoyant Plumes in Stratified Ambients

Abstract

Buoyant plumes in stratified ambients arise in wildfire smoke transport, ventilation, and pollutant dispersion, where many-query prediction requires models that stay accurate and stable as the dynamics transition from free rise to internal-wave trapping. Operator Inference (OpInf) learns polynomial reduced models from snapshots, and the Boussinesq plume depends affinely on the squared buoyancy frequency $N^2$. However, this compatibility masks two stability gaps: a single affine operator family can distort the per-parameter dynamics into unstable spectra, and for quadratic OpInf models, finite-time blowup is governed by a cubic energy injection channel from the inferred quadratic operator. To address these gaps, we develop cyclic-skew regime-bridged OpInf (CR-OpInf). We derive an affine-inadequacy diagnostic that links the departure from a shared affine family to destabilizing spectral drift before integration. In place of the shared affine fit, we interpolate per-parameter operators with a Gaussian kernel. We also extend the cyclic-skew quadratic constraint to the buoyancy-source-forced parametric setting, closing the cubic energy channel and isolating this closure as the boundedness mechanism. We further introduce a regime-bridging basis split that resolves free rise and trapped intrusion structures while preserving the cyclic-skew structure under convex interpolation. Applied to an unconfined Boussinesq plume and a viscous Burgers problem spanning a smooth-to-shock transition, CR-OpInf remains bounded for five training horizons, avoids the divergence of affine and unconstrained variants, tracks the plume rise, and recovers stratification-dependent rise scaling at moderate horizons. To our knowledge, this is the first parametric OpInf study of an unconfined stratified plume.

Braided endovascular implants for intracranial aneurysms: mechanics, hemodynamics, and clinical translation

Endovascular implants prevent intracranial aneurysm rupture by altering the mechanical and hemodynamic environment at the aneurysm neck. Yet many in silico workflows prescribe or reconstruct the post-deployment geometry before computing flow, leaving unresolved the mechanics that create the clinically relevant interface. Here we review braided intraluminal flow diverters, intrasaccular devices, and emerging flow-disruption concepts across deployment mechanics, inter-wire and wire-wall contact, superelasticity, wall apposition, pore geometry, computational fluid dynamics, and fluid-structure interaction. We connect these modeling choices to neck coverage, malapposition, migration, deformation, and durability, and distinguish established evidence from mechanistic inference and prospective hypotheses. We argue that model fidelity should match the clinical question: prescribed or fast placement may support screening, whereas questions of coverage, apposition, compaction, and migration benefit from mechanically plausible deployment states. An interface-resolved mechanics-to-flow framework, supported by measurable validation targets and standardized reporting, could improve device design and enable more reliable patient-specific treatment planning.

LAO+: Focused, Multi-Replica Accelerated Orthogonal Sampling for Robust Binding Free Energy Predictions

We introduce Lambda-ABF-OPES+ ($\text{LAO+}$), a multi-replica framework for absolute binding free energy (ABFE) calculations that accelerates both the alchemical transformation and the slow ``orthogonal'' motions coupled to it. At the core of $\text{LAO+}$ is a partitioned $λ$-dynamics scheme that concentrates sampling on the abrupt rehydration of the binding pocket, the main bottleneck of ligand decoupling, while Hamiltonian replica exchange relays configurations along the entire alchemical path. This drives a fourfold gain in convergence speed for the rate-limiting leg over our previous $\text{LAO}$ method, which was itself up to ninefold faster than Lambda-ABF, an overall gain of up to $\sim$36-fold. Applied to 28 protein--ligand complexes across five targets, $\text{LAO+}$ achieves a root-mean-square error of 0.83~kcal~mol$^{-1}$ and a correlation of 0.87 with experiment from only 63~ns of sampling per ligand. Moreover, dedicated reservoir replicas accelerate any slow degree of freedom that can be described by a collective variable. We demonstrate this by recovering hidden protein and ligand conformational states, for example in T4 lysozyme, BRD4 and JNK1. Together, these advances make accurate ABFE calculations practical for drug discovery.

A Scalable Sharp-Interface Immersed Boundary Method for Moving Boundary Flow Simulation on Multi-GPU Systems

Sharp-interface immersed boundary methods provide a versatile framework for simulating flows with complex moving boundaries, but their irregular data structures, dynamic sparsity, and discontinuous pressure operators present significant challenges for efficient multi-GPU implementation. This work presents a fully GPU-native, multi-GPU acceleration of the sharp-interface immersed boundary solver, ViCar3D. The implementation combines a hybrid OpenACC/CUDA programming model with a matrix decomposition that isolates dynamically changing immersed-boundary operators, GPU-optimized geometric kernels, asynchronous dual-stream sparse matrix--vector execution, and a symmetric-memory-based sparse communication strategy. A GPU-oriented iterative solver employing a preconditioned stabilized bi-conjugate gradient (BiCGStab) method is developed to efficiently treat the pressure Poisson equation on highly stretched Cartesian grids. The resulting framework preserves the accuracy and versatility of the original solver while delivering substantial acceleration and scalability for simulations involving complex moving and deforming immersed boundaries, thereby extending the applicability of sharp-interface immersed boundary methods to extreme-scale computational fluid dynamics.
